Hm, I'm sorry to say this, but I don't think so.
Most of my source files are just 256x, and there is one major problem: Minecraft doesn't supports Mip-Mapping, which will lead to a heavy Aliasing and pixelated blocks :(

I have a GTX 570 and can already get hundreds of frames per second running the 256x texture packs, any chance you can make a 512x512? ;)
